Two major highways run north and south through the heart of Pevely, Interstate Highway 55 and U.S. Highway 61/67. Both are intersected by State Highway Z, an east/west route serving as a collector for the two arteries and as a link to the Jefferson County Seat just 7 miles west. Interstate 55 links St. Louis to Chicago and points east as well to Memphis, Tennessee, and beyond to the south.

The I-55 corridor is the lifeline of the area and Pevely has commercial and industrial land available for development around the easy access interchange serving the City. In addition to the two major highways, two famous railroads travel through Pevely, the Burlington Northern Santa Fe and the Union Pacific. From Pevely, one can easily access a full range of transportation services to include Lambert International Airport in St. Louis, a major hub served by several airlines, as well as Mississippi River Port operations.

Pevely also has a Commuter Parking Lot located on Weier Road, just south of State Highway Z, and along Interstate Highway 55.
